Project summary: Undertook a project to characterize the 3D structure of mammalian coatomer complex I (COPI), which is involved in intra cellular trafficking; using a combination of biochemistry and single particle electron microscopy. The structural data gleaned from this study shed light on its function within the cell.
Duties assumed in this role: Characterized the 3D structure of the protein complex of COPI, using a combination of biochemistry and microscopy.
Established and implemented the protocols; wrote experimental guidelines and laboratory manuals pertinent to the project.
Trained and supervised undergraduate students.
Wrote a grant relevant to the project and secured a fellowship.
Presented talks related to the work at conferences and symposia.
Manuscript, intended for publication in peer reviewed journals in preparation.

PCR, DNA cloning and site-directed mutagenesis, and cell biology techniques – Sf9 cell culture, transfection, and generating competent cells.

Protein purification.
Biochemical characterization of protein and protein complexes by Western Blot, SDS-PAGE, Native-PAGE, Blue-Native PAGE, isoelectric focusing, and density gradient ultracentrifugation.

Negative stain electron microscopy. Random conical tilt (RCT), and working knowledge of cryo electron microscopy. Knowledge of electron microscopy image processing software like EMAN2, XMIPP, PRIME and RELION. Knowledge of molecular modeling and 3D visualization tools UCSF Chimera, Swiss PDB Viewer, Rasmol, VMD.
Familiar with data analysis packages like Origin and GraphPad.
Known computer languages: C, C++, currently learning Python for EM data analysis, with a strong interest in getting more experience in the computational analysis of EM data.
