Summary
Sodium tetrathionate is used to remove CoASH by oxidation so that the reaction is toward the right direction. Then, [14C]acetyl-carnitine is separated from [1-14C]acetyl-CoA by passing the mixture through an anion-exchange resin AG2-X8 (5g resin/100 mL sample). The isotope content in the elute ([14C]acetyl-carnitine) is determined by a scintillation counter.Further details
