Summary

A versatile technique for labeling and imaging cells in culture and in vivo is presented. A metabolically biotinylated reporter protein is expressed on the cell surface which can then be tracked with any imaging agent coupled to streptavidin. A model recombinant reporter protein incorporates the prokaryotic Propionibacterium shermanii 1.3S transcarboxylase domain biotin acceptor peptide (BAP) between an N-terminal signal sequence and the platelet derived growth factor receptor transmembrane domain (TM)1. The BAP sequence provides a means to track any protein and the BAP-TM reporter allows for non-invasive real time imaging of any cell type transduced to express it in culture or in vivo using a variety of techniques including optical1,2, magnetic resonance1,3 and positron emission tomogrophic imaging4.
