Abstract
Abdominal aortic aneurysm (AAA) is a life-threatening vascular condition marked by localized aortic dilation and structural weakening of the vessel wall, often leading to catastrophic complications such as rupture. As one of the leading causes of acute aortic-related mortality in the elderly, AAA rupture underscores the critical need for effective therapeutic strategies to prevent this fatal outcome. Current clinical management relies heavily on surgical or endovascular repair for larger aneurysms to reduce rupture risk; however, early diagnostic tools and pharmacological interventions remain limited. Advances in nanoscience present promising opportunities for targeted imaging and drug delivery in AAA treatment. Multifunctional nanoparticles (NPs) can be precisely engineered with ligands or biomembrane coatings to enable site-specific drug accumulation, minimizing systemic side effects. Beyond targeted delivery, NPs enhance drug solubility, prolong circulation half-life, improve bioavailability, and maximize therapeutic efficiency. Furthermore, carefully designed nanobiomaterials can maintain long-term stability within the vascular system without cellular internalization. This review highlights the applications of NPs in AAA imaging and therapy, summarizes recent progress in NPs-based diagnostics and treatments, and discusses their potential to transform clinical outcomes in AAA management.
