Abstract
The 80% ethanol extract of Chuanxiong Rhizoma from Guizhou was isolated and purified using various chromatographic techniques, including macroporous adsorption resin, silica gel, ODS, and Toyopearl HW-40F. The structures of the isolated compounds were elucidated by comprehensive spectroscopic methods. Twenty-two compounds were identified as(7R,7'R)1,1'-dibutyl-6,6',7'-tetrahydro-[1,1'-biisobenzofuran]-3,3'(1H,1'H)-dione(1),(3S)-butylphthalide(2), neocnidilide(3), chuanxiongdiodide A(4), senkyunolide J(5), trans-neocnidilide(6), levistolide A(7), riligustilide(8),(+)-dehydroabietane(9), albiflorin R_1(10), 7α,18-dihydroxydehydroabietanol(11), dehydroabietol(12),(4aS,4bR,6S)-7-(tert-butyl)-1,1,4a, 4b, 6-pentamethyl-1,2,3,4,4a, 4b, 5,6,10,10a-decahydrophenanthrene(13), 4-(4',4'-dimethyl-2'-vinyldecyl)-1,2-benzene-1,2-diol(14), β-sitosterol(15), β-stigmasterol(16), β-stigmasterol linoleate(17), homostigmast-5-en-3β-ol(18), 1,1'-oxybis(1,4)-di-tert-butylbenzene(19), 2,3-dihydroxypropyl(Z,Z)-9,12-octadecadienate(20), piperyamine A(21), and 13-isopropyl-pentacosane(22). Among them, compound 1 was identified as a new compound, while compounds 9, 11-14, and 18-22 were isolated from this plant for the first time. The results of in vitro activity screening indicated that compounds 11 and 20 exhibited significant anticoagulant activity, while compounds 2, 9, 14, and 19 showed good activity, and compounds 3, 5, 7-8, 12-13, 17, and 21 demonstrated moderate activity.
