Identification and characterization of the virome in cell-free DNA from peripheral blood of pregnant women in Shaanxi Province, China.

Abstract

This study characterizes the viral landscape identified within cell-free DNA (cfDNA) from peripheral blood samples of pregnant women in Shaanxi Province, China. Using non-invasive prenatal testing (NIPT) combined with high-throughput sequencing, we analyzed the virome composition in 20,035 maternal plasma samples to determine viral prevalence, regional variations, and potential associations with pregnancy outcomes. Seventy-one distinct viral species were detected, with Human Herpesvirus 7 (HHV-7) being the most common, identified in 9.41% of samples. The overall viral positivity rate among pregnant women in Shaanxi Province was 14.01%, significantly higher than the national average for China. Notable regional disparities in viral prevalence were observed among different cities within the province. Moreover, infection with certain viruses was potentially linked to adverse pregnancy outcomes, including birth defects. Our findings illustrate the distribution and prevalence of viral infections among pregnant women across Shaanxi Province and highlight the possible negative implications of specific viral infections for pregnancy outcomes. These results provide scientific evidence supporting region-specific viral monitoring and targeted public health interventions, particularly in areas exhibiting high infection rates, to minimize and manage risks associated with adverse pregnancy outcomes.
