Abstract

Objective: To analyze the molecular typing, drug resistance, and virulence factors of Salmonella isolates collected in Liaocheng City, Shandong Province, from 2023 to 2024. Methods: A total of 43 Salmonella strains were subjected to serotyping, antimicrobial susceptibility testing, and whole-genome sequencing (WGS). The Salmonella genomes were analyzed for multilocus sequence typing (MLST), core genome MLST (cgMLST), antimicrobial resistance genes, and virulence factors. Results: A total of 15 serotypes were identified among the 43 Salmonella strains, with the predominant serotype being Salmonella enteritidis (32.56%, 14/43). Sixteen sequence types (STs) were identified, and the dominant ST was ST11 (32.56%, 14/43). The cgMLST analysis showed that the 43 strains were genetically diverse and that strains with identical STs clustered closely together. The resistance rates to ampicillin, tetracycline, and ampicillin-sulbactam were 79.07% (34/43), 69.77% (30/43), and 53.49% (23/43), respectively. The multi-drug resistance rate reached 67.44% (29/43). Forty-seven antimicrobial resistance genes were detected. The aminoglycoside resistance gene aac(6')-Iaa was carried by all strains. The genes blaTEM-1B, tet(A), and sul2 also exhibited high carriage rates. Twenty Salmonella isolates carried mutations in the gyrA gene associated with increased fluoroquinolone resistance. The 43 Salmonella isolates carried 16 classes of virulence factors, with 100% carriage rates for virulence factors related to fimbrial adherence, macrophage-inducible genes, magnesium ion transport, non-fimbrial adherence, regulation, and the secretion system. Conclusion: Salmonella enteritidis and ST11 are the predominant serotype and sequence type in Liaocheng City. The isolates show severe multidrug resistance and carry abundant virulence factors.
