Bovine umbilical cord tissue‑derived MSCs and its conditioned medium promote the healing of excisional wound in rats.

Abstract

The goal of skin tissue regeneration is to achieve structural and functional reconstruction, reduce scar formation, and improve wound healing quality. The present study highlights the wound healing and tissue regeneration capability of bovine umbilical cord-derived mesenchymal stem cells (bUC-MSCs) and its cell culture supernatant, i.e. conditioned media (CM). The cells were isolated and characterized from umbilical tissue off-cut. Excisional wound model was created in adult rats. In separate groups the bUC-MSCs and reconstituted lyophilized-CM were injected subcutaneously around the wound periphery at a regular interval for 3 weeks. Significant improvement (p < 0.05) in wound contraction percentage was noticed in bUC-MSC and CM groups as early as day 2. The histo-morphological analysis revealed that in bUC-MSC and CM-treated groups enhanced monocytoid cell migration was noticed in inflammatory phase, exhibited superior collagen organization, and wound remodeling, which are critical for minimizing scar formation and ensuring functional recovery of the skin. The sustained effects observed after four weeks suggest that both bUC-MSCs and CM not only accelerate early healing processes but also contribute to the structural integrity and durability of the healed tissue. Overall, bUC-MSCs and its conditioned media represent a valuable addition to the toolkit of regenerative medicine.
