Abstract
The incorporation of silver nanoparticles and an efficient dissolving microarray patch (DMP) has been developed to overcome the limitation of the skin delivery procedure and enhance the penetration of antibacterial agents into infected skin. This study investigated the potential of green-synthesized silver nanoparticles (GAgNPs) as antibacterial agents against wound infections. The fabrication process of DMP incorporating GAgNPs is simple and suitable for large-scale production. The GAgNPs-DMP exhibits antibacterial activity against both Gram-positive and Gram-negative bacteria in a dose-dependent manner, with inhibition observed at approximately 106 CFU/mL. The wound-closure model demonstrated the biocompatibility and improved in vitro cell migration of the developed patches on HDF cells at an optimal dosage of 5.4 µg/mL. GAgNPs-DMP exhibited an initial rapid release phase followed by a slower release phase, with approximately 89% of total silver released by 8 h and a subsequent plateau indicating its potential as an effective drug delivery system for enhanced healing of infected wounds. This finding demonstrates the important biological function of GAgNPs, as evidenced by their antibacterial properties, whereas the polymer matrix is reportedly responsible for migration-related aspects. The synergistic effect of the GAgNPs-DMP presents a promising developed biopolymeric patch for transdermal drug delivery.
