Abstract
The escalating threat of multidrug-resistant (MDR) bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA), urgently demands integrated multimodal therapies. However, it remains challenging to develop all-in-one platforms that synchronize photothermal therapy (PTT), photodynamic therapy (PDT), and ion release under a single near-infrared (NIR) trigger. Herein, a van der Waals composite AgPor/Hj, assembled from Ag(II) porphyrin and graphene-TiO2 heterojunction, is reported. Synergistic hot hole injection from the heterojunction to the porphyrin macrocycle greatly enhances NIR absorption, achieving an ultrahigh photothermal conversion efficiency of 64.5% for porphyrin-based systems. Crucially, under a single 808 nm NIR irradiation, the engineered interface synchronizes PTT, PDT, and ultrafast Ag(II) release. This single-NIR-triggered triple-modal synergy enables outstanding biofilm eradication and potent MRSA inhibition both in vitro and in vivo. AgPor/Hj effectively eliminates bacteria in infected wounds and accelerates tissue repair, significantly outperforming vancomycin, the first-line antibiotic for MRSA. Combined with excellent biocompatibility, low cost, and easy preparation, this synergistic platform provides a highly promising strategy for treating MDR bacterial wound infections.
