Abstract

Triple-negative breast cancer (TNBC) is highly aggressive, with limited effective therapies and poor clinical outcomes. Here, we developed a dual RNAi prodrug with enhanced in vitro therapeutic efficacy by simultaneously silencing two overexpressed BCL2 and CDK1 genes associated with tumor survival and cell cycle progression pathways, respectively, in TNBC cells. Compared with conventional dual siRNA treatment, the dual RNAi prodrug exhibited efficient gene silencing and improved anticancer activity through coordinated suppression of survival and cell cycle pathways.
