Abstract

Monoclonal antibodies have served as research tools to characterize specific proteins and biomolecules to understand their role in biological processes and disease. However, because of nonspecific reactivity with other components in samples, specific reactivity can be difficult to interpret. Using web-based tools, standard hybridoma technology, and standard immunostaining methods, we describe the development of a tryptophan 2,3-dioxygenase 2 (TDO2)-targeted antibody with high specificity. In addition to providing a unique reagent needed as a research tool to investigate the role of TDO2 in preclinical models and human tissue samples, herein we provide methods to characterize TDO2 expression in human tissue samples and murine disease models. We note also that the general protocol described in this chapter will be useful to investigators interested in developing monoclonal antibodies against other targets of interest as well.
