Abstract

Objective: To investigate the effect of Doublecortin-like kinase 1 (DCLK1) on the angiogenic capacity of colon cancer SW480 and HCT116 cells and its underlying molecular mechanism. Methods: A pan-cancer analysis of DCLK1 was performed based on The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) databases. The correlations between DCLK1 expression level in colon cancer and tumor angiogenesis as well as patient prognosis were further analyzed. DCLK1-knockdown SW480 and DCLK1-overexpressing HCT116 cell lines were constructed via plasmid transient transfection. Protein microarray sequencing was used to detect changes in the expression of angiogenesis-related proteins and to conduct pathway analysis. A colon cancer tissue microarray[purchased from Shanghai Xinchao Biotechnology Co., Ltd. (Shanghai, China) in 2025] was subjected to multiplex fluorescent staining to observe the expression relationship between DCLK1 and vascular endothelial marker molecules. The effect of DCLK1 on angiogenic ability was evaluated through an angiogenesis assay. Results: Database analysis revealed that DCLK1 expression was upregulated in colon cancer and multiple other cancer types (all P<0.05). High DCLK1 expression was associated with poorer overall survival (HR=1.739, 95% CI: 1.051-2.876, P=0.029) and disease-specific survival (HR=2.842, 95% CI: 1.283-4.804, P=0.005) in patients. In colon cancer, patients with high DCLK1 expression had more advanced T stage (T3/4 vs. T1/2, P=0.011), more frequent lymph node metastasis (N1/2 vs. N0, P=0.011), and DCLK1 expression was significantly positively correlated with angiogenesis in colon cancer (r=0.459, P<0.001). DCLK1 expression levels showed significant positive correlations with vascular endothelial marker genes CD31, CD34, ENG, VWF, VEGFC, and VEGFR2 (all P<0.001). KEGG and Gene Set Enrichment Analysis (GSEA) showed that DCLK1 co-expressed genes were significantly enriched in pathways related to angiogenesis. Multiplex fluorescence staining of the tissue microarray demonstrated upregulated co-expression of DCLK1 and CD31 in colon cancer tissues. In vitro experiments confirmed that colon cancer cells overexpressing DCLK1 exhibited enhanced angiogenic capacity (P<0.001). Conclusion: DCLK1 promotes angiogenesis in colon cancer by regulating multiple molecules and pathways.
