Abstract

Calpains are calcium-activated cysteine proteases present in the cytoplasm and mitochondria that are essential to physiological processes; however, the functions of calpain-1 in the brain are not well understood. Therefore, we explored novel calpain-1 substrates using the GPS-CCD software package. Calpain inhibitors, calpeptin and Tat-μCL, suppressed cleavage of mitochondrial antiviral signaling protein (MAVS) c in the presence of exogenous calcium. Moreover, calpain-1 knockout mice suppressed MAVS cleavage in cerebral ischemia/reperfusion (I/R) injury. These findings indicate that MAVS is a novel calpain-1 substrate. As MAVS degradation prevents inflammation, its cleavage might contribute to the suppression of inflammation in I/R injury.
