Abstract

Muscle stem cells are essential for skeletal muscle regeneration, but their scarcity makes epigenetic studies technically and financially challenging. Here, we describe a robust chromatin immunoprecipitation (ChIP) protocol compatible with freshly isolated muscle stem cells using as few as 25,000 cells. This method avoids sample pooling, reduces animal use, and enables reliable analysis of chromatin modifications and transcription factor binding.
