Abstract

The rapid expansion of electrochemiluminescence (ECL) biosensing has created growing demand for efficient signal amplification strategies capable of meeting the sensitivity requirements of microRNA analysis. Among the available functional materials, metal-organic frameworks (MOFs) have evolved from simple signal carriers into multifunctional platforms that integrate probe loading, catalytic enhancement, programmable nucleic acid reactions, and emerging CRISPR-assisted amplification within a single sensing architecture. This review provides a mechanism-oriented perspective on MOF-assisted signal amplification strategies, highlighting how different amplification mechanisms govern analytical performance and practical applicability. Beyond summarizing representative biosensing systems, the advantages, limitations, and suitable application scenarios of different amplification strategies are critically compared to establish guidelines for rational biosensor design. Finally, the remaining obstacles to clinical translation are discussed, together with future opportunities for developing robust, integrated, and clinically deployable MOF-based ECL biosensing platforms.
